Genotyping of transgenic lines

Manager: Jean-Christophe Helbling

Operational team: Céline Ducroix-Crépy et Eva Bruchet

Goal

To ensure the reproduction, management and selection of transgenic lines in the Nutrineuro laboratory, using rapid and reliable genotyping techniques adapted to optimal colony management.

Our skills

  • Rapid DNA extraction: from mouse tail biopsies (5 mm).
  • Simplex and multiplex PCR techniques: for targeted and simultaneous amplification of several genetic markers.
  • Agarose gel electrophoresis: to visualize amplification results.
  • Hot Start Taq Pol polymerization enzyme: an innovation produced and purified directly by our team for increased reliability and efficiency.

Applications

The genotyping division offers specific PCRs for line discrimination and pre-birth sexing:

Line organization

  • Management and monitoring of transgenic lines using the ANISE interface, facilitating precise traceability of crosses and genotypes.

Equipment available

  • Applied Biosystems 2720 thermal cycler: for simplex and multiplex PCR.
  • Agilent thermocycler 8800 overcycler technology
  • Applied simpliAmp Applied biosystems thermocycler
  • Lifeeco Bioer thermal cycler
  • E-BOX imaging system, Vilber: for visualization and analysis of agarose gels.

Highlights

  • Fast, cost-effective approach to genotyping large colonies.
  • In-house innovation with Hot Start Taq Pol enzyme.
  • Optimized line management with dedicated tools, ANISE web interface.
